(The Decreased Serum Progesterone Concentrations and Extracellular Matrix and Cellular Components of Skin as Indicators of Aging in Rats)

ABSTRAK

Penelitian ini bertujuan mengetahui kadar kolagen kulit (matriks ekstraseluler), jumlah sel (kadar DNA), aktivitas sintetik (kadar RNA) pada jaringan kulit, serta kadar hormon progesteron pada berbagai tingkatan umur tikus, dan memperoleh umur tikus ovariektomi yang cocok digunakan sebagai hewan model penuaan. Rancangan percobaan yang digunakan adalah rancangan acak lengkap (RAL) yang terdiri atas sembilan kelompok percobaan masing-masing terdiri atas tiga ekor tikus, yaitu umur 12 bulan (K1), 18 bulan (K2), 24 bulan (K3), 30 bulan (K4), 36 bulan (K5), umur 12 bulan dalam kondisi 1 bulan pascaovariektomi (K6), 12 bulan dalam kondisi 3 bulan pascaovariektomi (K7), 18 bulan dalam kondisi 1 bulan pascaovariektomi (K8), dan umur 24 bulan dalam kondisi 1 bulan pascaovariektomi (K9). Data yang diperoleh dianalisis dengan menggunakan analisis varian dan dilanjutkan dengan uji Duncan. Komponen matriks ekstraseluler dan matriks seluler menurun seiring dengan bertambahnya usia. Umur tikus normal yang cocok digunakan sebagai hewan model penuaan adalah tikus umur 24-36 bulan. Tikus ovariektomi yang cocok digunakan sebagai hewan model penuaan adalah tikus umur 12 bulan pascaovariektomi 3 bulan.

ABSTRACT

This study was designed to determine the levels of skin collagen (extracellular matrix), the number cell (DNA concentration), synthetic activity (RNA concentration), and serum progesterone concentrations in various agesin rats, and to obtain suitable age of ovaries to rats used as animal models of aging. Experimental design used was Completely Randomized Design (CRD) consisted of 9 experimental groups, each consisted of 3 rats i.e., rats aged 12 months (K1), 18 months (K2), 24 months (K3), 30 months (K4) 36 months (K5), 12 months with one month post ovariectomy (K6), 12 months with 3 months post ovariectomy (K7), 18 months with one month post ovariectomy (K8), and 24 months with one month postovariectomy (K9). The data obtained were analyzed using Analysis of Variance (ANOVA) followed by Duncan test. Extracellular matrix component sand cellular matrix decreased within creasing of age. Ages of normal rats that were suitable to be used as animal models of aging were 24-36 months. Ovariectomy rats that were suitable to be used as animal models of aging were 12 months with three months postovariectomy.
